I'm quite a creature of habit, and I've always played rogue-likes with the keyboard. But it doesn't really seem practical to try to use entirely the keyboard. I've tried clicking all the options, and there are problems. I can't tell what I'm auto-picking up, for instance. It just goes right into the inventory. What about a "get" command? And ramming the chests and levers doesn't necessarily seem to open them even with "move as use." Am I missing something? Otherwise I might as well just go with all-mouse, but, *grump* *grump*
There are some options in the settings panel that will help set you up to use keyboard more primarily. The autoloot does need to have the item types to pick up selected in the autoloot option panel (though I'm thinking now that a global on/off would have been sufficient). Patch 1.0.6 is coming out in the next few days and it should enable some additional interactions using keyboard commands that'll make life easier.
On the topic of all-mouse, is there a way to rest using the mouse? I haven't found an on-screen button for it and I end up having to use space bar. If there isn't a way, I think one good place could be the portrait. Clicking on the portrait would cause you to rest a turn. So far, everything else other than resting seems to be do-able through mouse-only.
